Global change has been acknowledged as one of the main threats to the biosphere and its provision of ecosystem services, especially in marine ecosystems. Seagrasses play a critical ecological role in coastal ecosystems, but their responses to ocean acidification (OA) and climate change are not well understood. There have been previous studies focused on the effects of OA, but the outcome of interactions with co-factors predicted to alter during climate change still needs to be addressed. For example, the impact of higher CO2 and different hydrodynamic regimes on seagrass performance remains unknown. We studied the effects of OA under different current velocities on productivity of the seagrass Zostera noltei, using changes in dissolved oxygen as a proxy for the seagrass carbon metabolism, and release of dissolved organic carbon (DOC) in a four-week experiment using an open-water outdoor mesocosm. Under current pH conditions, increasing current velocity had a positive effect on productivity, but this depended on shoot density. However, this positive effect of current velocity disappeared under OA conditions. OA conditions led to a significant increase in gross production rate and respiration, suggesting that Z. noltei is carbon-limited under the current inorganic carbon concentration of seawater. In addition, an increase in non-structural carbohydrates was found, which may lead to better growing conditions and higher resilience in seagrasses subjected to environmental stress. Regarding DOC flux, a direct and positive relationship was found between current velocity and DOC release, both under current pH and OA conditions. We conclude that OA and high current velocity may lead to favourable growth scenarios for Z. noltei populations, increasing their productivity, non-structural carbohydrate concentrations and DOC release. Our results add new dimensions to predictions on how seagrass ecosystems will respond to climate change, with important implications for the resilience and conservation of these threatened ecosystems.

Short-term spatial and temporal carbonate chemistry variability in two contrasting seagrass meadows: implications for pH buffering capacities

It has been hypothesized that highly productive coastal ecosystems, such as seagrass meadows, could lead to the establishment of ocean acidification (OA) refugia, or areas of elevated pH and aragonite saturation state (Ωa) compared to source seawater. However, seagrass ecosystems experience extreme variability in carbonate chemistry across short temporal and small spatial scales, which could impact the pH buffering capacity of these potential refugia. Herein, short-term (hourly to diel) and small-scale (across 0.01–0.14 km2) spatiotemporal carbonate chemistry variability was assessed within two seagrass meadows in order to determine their short-term potential to elevate seawater pH relative to source seawater. Two locations at similar latitudes were chosen in order to compare systems dominated by coarse calcium carbonate (Bailey’s Bay, Bermuda) and muddy silicate (Mission Bay, CA, USA) sediments. In both systems, spatial variability of pH across the seagrass meadow at any given time was often greater than diel variability (e.g., the average range over 24 h) at any one site, with greater spatial variability occurring at low tide in Mission Bay. Mission Bay (spatial ΔpH = 0.08 ± 0.08; diel ΔpH = 0.12 ± 0.01; mean ± SD) had a greater average range in both temporal and spatial seawater chemistry than Bailey’s Bay (spatial ΔpH = 0.02 ± 0.01; diel ΔpH = 0.03 ± 0.00; mean ± SD). These differences were most likely due to a combination of slower currents, a larger tidal range, and more favorable weather conditions for photosynthesis (e.g., sunny with no rain) in Mission Bay. In both systems, there was a substantial amount of time (usually at night) when seawater pH within the seagrass beds was lower relative to the source seawater. Future studies aimed at assessing the potential of seagrass ecosystems to act as OA refugia for marine organisms need to account for the small-scale, high-frequency carbonate chemistry variability in both space and time, as this variability will impact where and when OA will be buffered or intensified.
Nutrient loading fosters seagrass productivity under ocean acidification

The effects of climate change are likely to be dependent on local settings. Nonetheless, the compounded effects of global and regional stressors remain poorly understood. Here, we used CO2 vents to assess how the effects of ocean acidification on the seagrass, Posidonia oceanica, and the associated epiphytic community can be modified by enhanced nutrient loading. P. oceanica at ambient and low pH sites was exposed to three nutrient levels for 16 months. The response of P. oceanica to experimental conditions was assessed by combining analyses of gene expression, plant growth, photosynthetic pigments and epiphyte loading. At low pH, nutrient addition fostered plant growth and the synthesis of photosynthetic pigments. Overexpression of nitrogen transporter genes following nutrient additions at low pH suggests enhanced nutrient uptake by the plant. In addition, enhanced nutrient levels reduced the expression of selected antioxidant genes in plants exposed to low pH and increased epiphyte cover at both ambient and low pH. Our results show that the effects of ocean acidification on P. oceanica depend upon local nutrient concentration. More generally, our findings suggest that taking into account local environmental settings will be crucial to advance our understanding of the effects of global stressors on marine systems.

Predicting adaptive fitness to any environment requires mechanistic understanding of environmental influence on metabolic networks that control energy assimilation, growth, and reproduction. Although the potential impacts of environment on gene products are myriad, important phenotypic responses are often regulated by a few key points in metabolic networks where externally supplied resources or physiological reaction substrates limit reaction kinetics. Environmental resources commonly limiting seagrass productivity, survival, and growth include light and CO2 availability that control carbon assimilation and sucrose formation. Phosphate availability can also be important in oligotrophic tropical environments, particularly in the presence of carbonate sediments. Temperature and macronutrient oversupply (eutrophication) can act as confounding stressors, particularly in temperate environments. Photoacclimation can be regulated by electron transport pathways residing in the chloroplast stroma, but stress responses are often manifest by the expression of generalized stress response proteins, both of which appear to be affected by temperature and CO2 availability. A systems approach is employed to explore (1) the responses of seagrasses to the combined impacts of environmental limiting factors that control fundamental physiological processes leading to whole-plant performance; (2) sediment diagenetic processes that facilitate nutrient remineralization, carbon sequestration, and toxin neutralization; (3) interactions with other organisms induced by trophic cascades; and (4) impacts of human-induced climate change that affect system dynamics at numerous points in the network.
Global and local disturbances interact to modify seagrass palatability

Global change, such as warming and ocean acidification, and local anthropogenic disturbances, such as eutrophication, can have profound impacts on marine organisms. However, we are far from being able to predict the outcome of multiple interacting disturbances on seagrass communities. Herbivores are key in determining plant community structure and the transfer of energy up the food web. Global and local disturbances may alter the ecological role of herbivory by modifying leaf palatability (i.e. leaf traits) and consequently, the feeding patterns of herbivores. This study evaluates the main and interactive effects of factors related to global change (i.e. elevated temperature, lower pH levels and associated ocean acidification) and local disturbance (i.e. eutrophication through ammonium enrichment) on a broad spectrum of leaf traits using the temperate seagrass Cymodocea nodosa, including structural, nutritional, biomechanical and chemical traits. The effect of these traits on the consumption rates of the generalist herbivore Paracentrotus lividus (purple sea urchin) is evaluated. The three disturbances of warming, low pH level and eutrophication, alone and in combination, increased the consumption rate of seagrass by modifying all leaf traits. Leaf nutritional quality, measured as nitrogen content, was positively correlated to consumption rate. In contrast, a negative correlation was found between feeding decisions by sea urchins and structural, biomechanical and chemical leaf traits. In addition, a notable accomplishment of this work is the identification of phenolic compounds not previously reported for C. nodosa. Our results suggest that global and local disturbances may trigger a major shift in the herbivory of seagrass communities, with important implications for the resilience of seagrass ecosystems.

Macroalgae may mitigate ocean acidification effects on mussel calcification by increasing pH and its fluctuations

Ocean acidification (OA) is generally assumed to negatively impact calcification rates of marine organisms. At a local scale however, biological activity of macrophytes may generate pH fluctuations with rates of change that are orders of magnitude larger than the long-term trend predicted for the open ocean. These fluctuations may in turn impact benthic calcifiers in the vicinity. Combining laboratory, mesocosm and field studies, such interactions between OA, the brown alga Fucus vesiculosus, the sea grass Zostera marina and the blue mussel Mytilus edulis were investigated at spatial scales from decimetres to 100s of meters in the western Baltic. Macrophytes increased the overall mean pH of the habitat by up to 0.3 units relative to macrophyte-free, but otherwise similar, habitats and imposed diurnal pH fluctuations with amplitudes ranging from 0.3 to more than 1 pH unit. These amplitudes and their impact on mussel calcification tended to increase with increasing macrophyte biomass to bulk water ratio. At the laboratory and mesocosm scales, biogenic pH fluctuations allowed mussels to maintain calcification even under acidified conditions by shifting most of their calcification activity into the daytime when biogenic fluctuations caused by macrophyte activity offered temporal refuge from OA stress. In natural habitats with a low biomass to water body ratio, the impact of biogenic pH fluctuations on mean calcification rates of M. edulis was less pronounced. Thus, in dense algae or seagrass habitats, macrophytes may mitigate OA impact on mussel calcification by raising mean pH and providing temporal refuge from acidification stress.
Response of Posidonia oceanica seagrass and its epibiont communities to ocean acidification

The unprecedented rate of CO2 increase in our atmosphere and subsequent ocean acidification (OA) threatens coastal ecosystems. To forecast the functioning of coastal seagrass ecosystems in acidified oceans, more knowledge on the long-term adaptive capacities of seagrass species and their epibionts is needed. Therefore we studied morphological characteristics of Posidonia oceanica and the structure of its epibiont communities at a Mediterranean volcanic CO2 vent off Panarea Island (Italy) and performed a laboratory experiment to test the effect of OA on P. oceanica photosynthesis and its potential buffering capacity. At the study site east of Basiluzzo Islet, venting of CO2 gas was controlled by tides, resulting in an average pH difference of 0.1 between the vent and reference site. P. oceanicashoot and leaf density was unaffected by these levels of OA, although shorter leaves at the vent site suggest increased susceptibility to erosion, potentially by herbivores. The community of sessile epibionts differed in composition and was characterized by a higher species richness at the vent site, though net epiphytic calcium carbonate concentration was similar. These findings suggest a higher ecosystem complexity at the vent site, which may have facilitated the higher diversity of copepods in the otherwise unaffected motile epibiont community. In the laboratory experiment, P. oceanica photosynthesis increased with decreasing pHT (7.6, 6.6, 5.5), which induced an elevated pH at the leaf surfaces of up to 0.5 units compared to the ambient seawater pHT of 6.6. This suggests a temporary pH buffering in the diffusive boundary layer of leaves, which could be favorable for epibiont organisms. The results of this multispecies study contribute to understanding community-level responses and underlying processes in long-term acidified conditions. Increased replication and monitoring of physico-chemical parameters on an annual scale are, however, recommended to assure that the biological responses observed during a short period reflect long-term dynamics of these parameters.
Unexpected resilience of a seagrass system exposed to global stressors

Despite a growing interest in identifying tipping points in response to environmental change, our understanding of the ecological mechanisms underlying non-linear ecosystem dynamics is limited. Ecosystems governed by strong species interactions can provide important insight into how non-linear relationships between organisms and their environment propagate through ecosystems, and the potential for environmentally mediated species interactions to drive or protect against sudden ecosystem shifts. Here, we experimentally determine the functional relationships (i.e., the shapes of the relationships between predictor and response variables) of a seagrass assemblage with well-defined species interactions to ocean acidification (enrichment of CO2) in isolation and in combination with nutrient loading. We demonstrate that the effect of ocean acidification on grazer biomass (Phyllaplysia taylori and Idotea resecata) was quadratic, with the peak of grazer biomass at mid-pH levels. Algal grazing was negatively affected by nutrients, potentially due to low grazer affinity for macroalgae (Ulva intestinalis), as recruitment of both macroalgae and diatoms were favored in elevated nutrient conditions. This led to an exponential increase in macroalgal and epiphyte biomass with ocean acidification, regardless of nutrient concentration. When left unchecked algae can cause declines in seagrass productivity and persistence through shading and competition. Despite quadratic and exponential functional relationships to stressors that could cause a non-linear decrease in seagrass biomass, productivity of our model seagrass – the eelgrass (Zostera marina)- remained highly resilient to increasing acidification. These results suggest that important species interactions governing ecosystem dynamics may shift with environmental change, and ecosystem state may be decoupled from ecological responses at lower levels of organization.

Moderate increase in TCO2 enhances photosynthesis of seagrass Zostera japonica, but not Zostera marina: implications for acidification mitigation

Photosynthesis and respiration are vital biological processes that shape the diurnal variability of carbonate chemistry in nearshore waters, presumably ameliorating (daytime) or exacerbating (nighttime) short-term acidification events, which are expected to increase in severity with ocean acidification (OA). Biogenic habitats such as seagrass beds have the capacity to reduce CO2 concentration and potentially provide refugia from OA. Further, some seagrasses have been shown to increase their photosynthetic rate in response to enriched total CO2 (TCO2). Therefore, the ability of seagrass to mitigate OA may increase as concentrations of TCO2 increase. In this study, we exposed native Zostera marina and non-native Zostera japonica seagrasses from Padilla Bay, WA (USA) to various levels of irradiance and TCO2. Our results indicate that the average maximum net photosynthetic rate (Pmax) for Z. japonica as a function of irradiance and TCO2 was 3x greater than Z. marina when standardized to chlorophyll (360 ± 33 μmol TCO2 mg chl−1 h−1 and 113 ± 10 μmol TCO2 mg chl−1 h−1, respectively). Additionally, Z. japonica increased its Pmax ~50% when TCO2 increased from ~1,770 to 2,051 μmol TCO2 kg−1. In contrast, Z. marina did not display an increase in Pmax with higher TCO2, possibly due to the variance of photosynthetic rates at saturating irradiance within TCO2 treatments (coefficient of variation: 30–60%) relative to the range of TCO2 tested. Our results suggest that Z. japonica can affect the OA mitigation potential of seagrass beds, and its contribution may increase relative to Z. marina as oceanic TCO2 rises. Further, we extended our empirical results to incorporate various biomass to water volume ratios in order to conceptualize how these additional attributes affect changes in carbonate chemistry. Estimates show that the change in TCO2 via photosynthetic carbon uptake as modeled in this study can produce positive diurnal changes in pH and aragonite saturation state that are on the same order of magnitude as those estimated for whole seagrass systems. Based on our results, we predict that seagrasses Z. marina and Z. japonica both have the potential to produce short-term changes in carbonate chemistry, thus offsetting anthropogenic acidification when irradiance is saturating.
Carbon and nitrogen allocation strategy in Posidonia oceanica is altered by seawater acidification

Rising atmospheric CO2 causes ocean acidification that represents one of the major ecological threats for marine biota. We tested the hypothesis that long-term exposure to increased CO2 level and acidification in a natural CO2 vent system alters carbon (C) and nitrogen (N) metabolism in Posidonia oceanica L. (Delile), affecting its resilience, or capability to restore the physiological homeostasis, and the nutritional quality of organic matter available for grazers. Seawater acidification decreased the C to N ratio in P. oceanica tissues and increased grazing rate, shoot density, leaf proteins and asparagine accumulation in rhizomes, while the maximum photochemical efficiency of photosystem II was unaffected. The 13C-dilution in both structural and non-structural C metabolites in the acidified site indicated quali-quantitative changes of C source and/or increased isotopic fractionation during C uptake and carboxylation associated with the higher CO2 level. The decreased C:N ratio in the acidified site suggests an increased N availability, leading to a greater storage of 15N-enriched compounds in rhizomes. The amount of the more dynamic C storage form, sucrose, decreased in rhizomes of the acidified site in response to the enhanced energy demand due to higher shoot recruitment and N compound synthesis, without affecting starch reserves. The ability to modulate the balance between stable and dynamic C reserves could represent a key ecophysiological mechanism for P. oceanica resilience under environmental perturbation. Finally, alteration in C and N dynamics promoted a positive contribution of this seagrass to the local food web.
Genome-wide transcriptional reprogramming in the seagrass Cymodocea nodosa under experimental ocean acidification

Here we report the first use of massive scale RNA-Sequencing to explore seagrass response to CO2-driven ocean acidification (OA). Large-scale gene expression changes in the seagrass Cymodocea nodosa occurred at CO2 levels projected by the end of the century. C. nodosatranscriptome was obtained using Illumina RNA-Seq technology and de novo assembly, and differential gene expression was explored in plants exposed to short-term high CO2 / low pH conditions. At high pCO2, there was a significant increased expression of transcripts associated to photosynthesis, including light reaction functions and CO2 fixation, and also to respiratory pathways, specifically for enzymes involved in glycolysis, in the tricarboxylic acid cycle and in the energy metabolism of the mitochondrial electron transport. The up-regulation of respiratory metabolism is probably supported by the increased availability of photosynthates and increased energy demand for biosynthesis and stress-related processes under elevated CO2 and low pH. The up-regulation of several chaperones resembling heat stress-induced changes in gene expression, highlighted the positive role these proteins play in tolerance to intracellular acid stress in seagrasses. OA further modifies C. nodosa secondary metabolism inducing the transcription of enzymes related to carbon-based-secondary compounds biosynthesis, in particular the synthesis of polyphenols and isoprenoid compounds that have a variety of biological functions including plant defense. By demonstrating which physiological processes are most sensitive to OA, this research provide a major advance in the understanding of seagrass metabolism in the context of altered seawater chemistry from global climate change.
Short-term growth and biomechanical responses of the temperate seagrass Cymodocea nodosa to CO2 enrichment

Seagrasses are often regarded as climate change ‘winners’ because they exhibit higher rates of photosynthesis, carbon fixation and growth when exposed to increasing levels of ocean acidification. However, questions remain whether such growth enhancement compromises the biomechanical properties of the plants, altering their vulnerability to structural damage and leaf loss. Here, we investigated the short-term (6 wk) effects of decreasing pH by CO2 enrichment on the growth, morphology and leaf-breaking force of the temperate seagrass Cymodocea nodosa. We found that the plant biomass balance under levels of acidification representative of short-term climate change projections (pH 8.04) was positive and led to an increase in leaf abundance in the shoots. However, we also found that plant biomass balance was negative under levels of acidification experienced presently (pH 8.29) and those projected over the long-term (pH 7.82). Leaf morphology (mean leaf length, thickness and width) was invariant across our imposed acidification gradient, although leaves were slightly stronger under [CO2] representative of short-term climate change. Taken together, these findings indicate that a subtle increase in growth and mechanical resistance of C. nodosa is likely to occur following short- to medium-term changes in ocean chemistry, but that these positive effects are unlikely to be maintained over the longer term. Our study emphasises the need to account for the interdependencies between environmental conditions and variations in multiple aspects of the structure and functioning of seagrass communities when considering the likely consequences of climate change.
Altered epiphyte community and sea urchin diet in Posidonia oceanica meadows in the vicinity of submarine volcanic CO2 vents

Ocean acidification (OA) predicted for 2100 is expected to shift seagrass epiphyte communities towards the dominance of more tolerant non-calcifying taxa. However, little is known about the indirect effects of such changes on food provision to key seagrass consumers. We found that epiphyte communities of the seagrass Posidonia oceanica in two naturally acidified sites (i.e. north and south sides of a volcanic CO2 vent) and in a control site away from the vent at the Ischia Island (NW Mediterranean Sea) significantly differed in composition and abundance. Such differences involved a higher abundance of non-calcareous crustose brown algae and a decline of calcifying polychaetes in both acidified sites. A lower epiphytic abundance of crustose coralline algae occurred only in the south side of the vents, thus suggesting that OA may alter epiphyte assemblages in different ways due to interaction with local factors such as differential fish herbivory or hydrodynamics. The OA effects on food items (seagrass, epiphytes, and algae) indirectly propagated into food provision to the sea urchin Paracentrotus lividus, as reflected by a reduced P. oceanica exploitation (i.e. less seagrass and calcareous epiphytes in the diet) in favour of non-calcareous green algae in both vent sites. In contrast, we detected no difference close and outside the vents neither in the composition of sea urchin diet nor in the total abundance of calcareous versus non-calcareous taxa. More research, under realistic scenarios of predicted pH reduction (i.e. ≤ 0.32 units of pH by 2100), is still necessary to better understand cascading effects of this altered urchin exploitation of food resources under acidified conditions on ecosystem diversity and function.
Effects of in situ CO2 enrichment on Posidonia oceanica epiphytic community composition and mineralogy

Alterations in seagrass epiphytic communities are expected under future ocean acidification conditions, yet this hypothesis has been little tested in situ. A Free Ocean Carbon Dioxide Enrichment system was used to lower pH by a ~0.3 unit offset within a partially enclosed portion (1.7 m3) of a Posidonia oceanica meadow (11 m depth) between June 21 and November 3, 2014. Leaf epiphytic community composition (% cover) and bulk epiphytic mineralogy were compared every 4 weeks within three treatments, located in the same meadow: a pH-manipulated (experimental enclosure) and a control enclosure, as well as a nearby ambient area. Percent coverage of invertebrate calcifiers and crustose coralline algae (CCA) did not appear to be affected by the lowered pH. Furthermore, fleshy algae did not proliferate at lowered pH. Only Foraminifera, which covered less than 3% of leaf surfaces, declined in manner consistent with ocean acidification predictions. Bulk epiphytic magnesium carbonate composition was similar between treatments and percentage of magnesium appeared to increase from summer to autumn. CCA did not exhibit any visible skeleton dissolution or mineral alteration at lowered pH and carbonate saturation state. Negative impacts from ocean acidification on P. oceanica epiphytic communities were smaller than expected. Epiphytic calcifiers were possibly protected from the pH treatment due to host plant photosynthesis inside the enclosure where water flow is slowed. The more positive outcome than expected suggests that calcareous members of epiphytic communities may find refuge in some conditions and be resilient to environmentally relevant changes in carbonate chemistry.

Agardh and Turbinaria conoides (J. Agardh) Kützing, were incubated in seawater of pH 8.0, 7.8 (ambient), 7.6, 7.4 and 7.2, to study the effects of changing seawater pH on halocarbon emissions. Eight halocarbon species known to be emitted by seaweeds were investigated: bromoform (CHBr3), dibromomethane (CH2Br2), iodomethane (CH3I), diiodomethane (CH2I2), bromoiodomethane (CH2BrI), bromochloromethane (CH2BrCl), bromodichloromethane (CHBrCl2), and dibromochloromethane (CHBr2Cl). These very short-lived halocarbon gases are believed to contribute to stratospheric halogen concentrations if released in the tropics. It was observed that the seaweeds emit all eight halocarbons assayed, with the exception of K. alvarezii and S. binderi for CH2I2 and CH3I respectively, which were not measurable at the achievable limit of detection. The effect of pH on halocarbon emission by the seaweeds was shown to be species-specific and compound specific. The highest percentage changes in emissions for the halocarbons of interest were observed at the lower pH levels of 7.2 and 7.4 especially in Padina australis and Sargassum spp., showing that lower seawater pH causes elevated emissions of some halocarbon compounds. In general the seaweed least affected by pH change in terms of types of halocarbon emission, was P. australis. The commercially farmed seaweed K. alvarezii was very sensitive to pH change as shown by the high increases in most of the compounds in all pH levels relative to ambient. In terms of percentage decrease in maximum quantum yield of photosynthesis (Fv∕Fm) prior to and after incubation, there were no significant correlations with the various pH levels tested for all seaweeds. The correlation between percentage decrease in the maximum quantum yield of photosynthesis (Fv∕Fm) and halocarbon emission rates, was significant only for CH2BrCl emission by P. australis (r = 0.47; p ≤ 0.04), implying that photosynthesis may not be closely linked to halocarbon emissions by the seaweeds studied. Bromine was the largest contributor to the total mass of halogen emitted for all the seaweeds at all pH. The highest total amount of bromine emitted by K. alvarezii (an average of 98% of total mass of halogens) and the increase in the total amount of chlorine with decreasing seawater pH fuels concern for the expanding seaweed farming activities in the ASEAN region.
Experimental impacts of climate warming and ocean carbonation on eelgrass Zostera marina

CO2 is a critical and potentially limiting substrate for photosynthesis of both terrestrial and aquatic ecosystems. In addition to being a climate-warming greenhouse gas, increasing concentrations of CO2 will dissolve in the oceans, eliciting both negative and positive responses among organisms in a process commonly known as ocean acidification. The dissolution of CO2 into ocean surface waters, however, also increases its availability for photosynthesis, to which the highly successful, and ecologically important, seagrasses respond positively. Thus, the process might be more accurately characterized as ocean carbonation. This experiment demonstrated that CO2 stimulation of primary production enhances the summertime survival, growth, and proliferation of perennial eelgrass Zostera marina from the Chesapeake region, which is regularly impacted by summer heat stress. The experiment also quantified the logarithmic response to CO2 in terms of shoot proliferation, size, growth and sugar accumulation that was fundamentally consistent with model predictions based on metabolic carbon balance derived from short-term laboratory experiments performed with other eelgrass populations from cool ocean climates and other seagrass species from tropical and temperate environments. Rather than acting in a neutral fashion or as an independent stressor, increased CO2 availability can serve as a quantitative antagonist to counter the negative impact of climate warming on seagrass growth and survival. These results reinforce the emerging paradigm that seagrasses are likely to benefit significantly from a high-CO2 world.
The influence of CO2 enrichment on net photosynthesis of seagrass Zostera marina in a brackish water environment

Seagrasses are distributed across the globe and their communities may play key roles in the coastal ecosystems. Seagrass meadows are expected to benefit from the increased carbon availability which might be used in photosynthesis in a future high CO2 world. The main aim of this study was to examine the effect of elevated pCO2 on the net photosynthesis of seagrass Zostera marina in a brackish water environment. The short-term mesocosm experiments were conducted in Kõiguste Bay (northern part of Gulf of Riga, the Baltic Sea) in June–July 2013 and 2014. As the levels of pCO2 naturally range from ca. 150 μatm to well above 1000 μatm under summer conditions in Kõiguste Bay we chose to operate in mesocosms with the pCO2 levels of ca. 2000, ca. 1000, and ca. 200 μatm. Additionally, in 2014 the photosynthesis of Z. marina was measured outside of the mesocosm in the natural conditions. In the shallow coastal Baltic Sea seagrass Z. marina lives in a highly variable environment due to seasonality and rapid changes in meteorological conditions. This was demonstrated by the remarkable differences in water temperatures between experimental years of ca. 8°C. Thus, the current study also investigated the effect of elevated pCO2 in combination with short-term natural fluctuations of environmental factors, i.e., temperature and PAR on the photosynthesis of Z. marina. Our results show that elevated pCO2 alone did not enhance the photosynthesis of the seagrass. The photosynthetic response of Z. marina to CO2 enrichment was affected by changes in water temperature and light availability.
Linking gene expression to productivity to unravel long- and short-term responses of seagrasses exposed to CO2 in volcanic vents

Ocean acidification is a major threat for marine life but seagrasses are expected to benefit from high CO2. In situ (long-term) and transplanted (short-term) plant incubations of the seagrass Cymodocea nodosa were performed near and away the influence of volcanic CO2 vents at Vulcano Island to test the hypothesis of beneficial effects of CO2 on plant productivity. We relate, for the first time, the expression of photosynthetic, antioxidant and metal detoxification-related genes to net plant productivity (NPP). Results revealed a consistent pattern between gene expression and productivity indicating water origin as the main source of variability. However, the hypothesised beneficial effect of high CO2 around vents was not supported. We observed a consistent long- and short-term pattern of gene down-regulation and 2.5-fold NPP decrease in plants incubated in water from the vents and a generalized up-regulation and NPP increase in plants from the vent site incubated with water from the Reference site. Contrastingly, NPP of specimens experimentally exposed to a CO2 range significantly correlated with CO2 availability. The down-regulation of metal-related genes in C. nodosa leaves exposed to water from the venting site suggests that other factors than heavy metals, may be at play at Vulcano confounding the CO2 effects.
Light availability and temperature, not increased CO2, will structure future meadows of Posidonia oceanica

We evaluated the photosynthetic performance of Posidonia oceanica during short-term laboratory exposures to ambient and elevated temperatures (24–25 °C and 29–30 °C) warming and pCO2 (380, 750 and 1000 ppm pCO2) under normal and low light conditions (200 and 40 μmol photons m−2 s−1 respectively). Plant growth was measured at the low light regime and showed a negative response to warming. Light was a critical factor for photosynthetic performance, although we found no evidence of compensation of photosynthetic quantum efficiency in high light. Relative Electron Rate Transport (rETRmax) was higher in plants incubated in high light, but not affected by pCO2 or temperature. The saturation irradiance (Ik) was negatively affected by temperature. We conclude that elevated CO2 does not enhance photosynthetic activity and growth, in the short term for P. oceanica, while temperature has a direct negative effect on growth. Low light availability also negatively affected photosynthetic performance during the short experimental period examined here. Therefore increasing concentrations of CO2 may not compensate for predicted future conditions of warmer water and higher turbidity for seagrass meadows.
Seagrass ecophysiological performance under ocean warming and acidification

Seagrasses play an essential ecological role within coastal habitats and their worldwide population decline has been linked to different types of anthropogenic forces. We investigated, for the first time, the combined effects of future ocean warming and acidification on fundamental biological processes of Zostera noltii, including shoot density, leaf coloration, photophysiology (electron transport rate, ETR; maximum PSII quantum yield, Fv/Fm) and photosynthetic pigments. Shoot density was severely affected under warming conditions, with a concomitant increase in the frequency of brownish colored leaves (seagrass die-off). Warming was responsible for a significant decrease in ETR and Fv/Fm (particularly under control pH conditions), while promoting the highest ETR variability (among experimental treatments). Warming also elicited a significant increase in pheophytin and carotenoid levels, alongside an increase in carotenoid/chlorophyll ratio and De-Epoxidation State (DES). Acidification significantly affected photosynthetic pigments content (antheraxanthin, β-carotene, violaxanthin and zeaxanthin), with a significant decrease being recorded under the warming scenario. No significant interaction between ocean acidification and warming was observed. Our findings suggest that future ocean warming will be a foremost determinant stressor influencing Z. noltii survival and physiological performance. Additionally, acidification conditions to occur in the future will be unable to counteract deleterious effects posed by ocean warming.
